This guide looks absolutely incredible and well made! People underestimate Li Li far to much. I think this guide will change that and I can finally start getting some good healers ;)
I agree that Two For One is quite good, but I think you might be overvaluing its output just a bit. Normally we get 1 heal every 3 seconds, or .333 heals per second. With Two For One we get 2 heals every 4.5 seconds, or .444 heals per second. That's only a 33% increase to our net output on heals, which hardly qualifies as doubling our overall healing.
Great guide, this build is fairly similar to what I usually run. I have a few comments though:
1) The Serpent definitely attacks chests at the moment, which allows Li Li and her allies to pick them up on Blackheart Bay really quickly.
2) On level 16, I feel Two for One is a huge trade-off. Having played with both it and Herbal Cleanse I'm still torn between the two. Two for One is definitely better for team fights, but I had so many situations when someone on my team got CC'd by the enemy and couldn't disengage properly. In cases like this I couldn't possibly save them with Two for One due to the increased cooldown on the Brew. Kung Fu Hustle would help of course, but it also helps with the base cooldown.
Similarly, when chasing down someone Herbal Cleanse's movement speed buff was often really handy. It's all about team comp of course and positioning, but I think it's really important to recognize that Two for One makes you much less effective at healing a target that's being focused and choosing it can easily come back to bite you.
3) While I think the Serpent could use a buff I also feel you're selling it a little short as a healing tool (its damage is laughable and uncontrollable anyway). First of all, it can become a targetable heal, something that Li Li lacks otherwise. Also, it's another heal beside your Brew, that works while you're casting the Jug ulti as well. For sure it's not amazing, but at least in Quick Matches it's workable. I often used it to heal someone clearing a merc camp while I was needed elsewhere for example (globes aren't everything :)). With Surging Winds it can become a respectable HoT in my opinion. Losing out on Shrink Ray can really hurt though, but depending on your team it's not the end of the world.
I agree with everything in this build. I do hate the increased cool down time caused from Two For One, but there isn't much in that tier I like more so I take it anyways, and then Kung Fu Hustle circumvents that negative, just gotta tough it out for 4 levels.
I now always take Mass Vortex, ever since I found out that increased ability power from Surging Winds applies to Healing Brew's heals as well. At first I thought it only applied to damage, but being applied to heals as well? I found Lili really needed this buff to keep up to healers like Rehgar and Uther.
All in all, good guide, good reasoning on your talent picks.
